產(chǎn)品概述
IFN 最初是一類哺乳動(dòng)物的細(xì)胞因子,它的特點(diǎn)是可以抑制病毒感染。除了抗病毒特性,其也具有抗增殖、參與免疫調(diào)節(jié)等其他功能。當(dāng)IFN與它的同源受體作用時(shí),信號(hào)快速在細(xì)胞內(nèi)傳遞,緊接著就出現(xiàn)抗病毒的狀態(tài)。干擾素-α(IFN-α),也被稱為白細(xì)胞干擾素,代表著一類相關(guān)但蛋白質(zhì)組成又不同的因子,他們具有大約 95%的氨基酸序列同源性且屬于I型IFN,都具有兩個(gè)亞基組成的細(xì)胞表面受體。I型IFN引起的主要的信號(hào)傳導(dǎo)通路是JAK1-STAT通路。
產(chǎn)品原理
本實(shí)驗(yàn)采用雙抗體夾心ELISA法??谷薎FN-α單抗包被于酶標(biāo)板上,同時(shí)加入標(biāo)本、標(biāo)準(zhǔn)品和生物素化的抗人IFN-α 抗體,標(biāo)本、標(biāo)準(zhǔn)品中的IFN-α 會(huì)與加入于孔中的生物素化的抗人IFN-α 抗體及包被于酶標(biāo)板上的單抗結(jié)合,形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標(biāo)記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有IFN-α,辣根過氧化物酶會(huì)使無色的顯色劑變藍(lán),加終止液變黃。在450nm 處測(cè)OD值,IFN-α濃度與OD450值之間呈正相關(guān),可通過繪制標(biāo)準(zhǔn)曲線求出標(biāo)本中IFN-α濃度。
