肝細(xì)胞生長(zhǎng)因子(Hepatocyte growth factor,HGF)是一種多效應(yīng)生長(zhǎng)因子,又稱(chēng)擴(kuò)散因子(scatter factor,SF)、肝細(xì)胞生成素A(hepatopoietin A)(肝細(xì)胞生成素B可能是A的片段)和乳腺生長(zhǎng)因子,最初從大鼠的血小板中純化獲得。成熟的肝細(xì)胞生長(zhǎng)因子是一個(gè)分子量82kDa,含674個(gè)氨基酸殘基的異二聚體糖蛋白,它是一類(lèi)因子小家族的一員,該家族還包括肝細(xì)胞生長(zhǎng)因子樣蛋白(也叫巨噬細(xì)胞刺激蛋白,MSP),這類(lèi)因子與其他已知的生長(zhǎng)因子幾乎沒(méi)有明顯的同源性。肝細(xì)胞生長(zhǎng)因子對(duì)肝細(xì)胞和其他上皮細(xì)胞,以及內(nèi)皮細(xì)胞、間充質(zhì)細(xì)胞和造血細(xì)胞類(lèi)具有多種不同的作用,包括促進(jìn)有絲分裂,形態(tài)形成和促使細(xì)胞運(yùn)動(dòng)等。
產(chǎn)品原理
本實(shí)驗(yàn)采用雙抗體夾心ELISA法??谷薍GF單抗包被于酶標(biāo)板上,標(biāo)本和標(biāo)準(zhǔn)品中的HGF會(huì)與單抗結(jié)合形成免疫復(fù)合物,游離的成分被洗去;加入生物素化的抗人HGF抗體,生物素化抗人HGF抗體與酶標(biāo)板上結(jié)合的標(biāo)準(zhǔn)品或樣本中的HGF結(jié)合而形成免疫復(fù)合物,游離的成分被洗去;加入辣根過(guò)氧化物酶標(biāo)記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有HGF,辣根過(guò)氧化物酶會(huì)使無(wú)色的顯色劑變藍(lán),加終止液變黃。在450nm處測(cè)OD值,HGF濃度與OD450值之間呈正相關(guān),可通過(guò)繪制標(biāo)準(zhǔn)曲線求出標(biāo)本中HGF濃度。
