產(chǎn)品概述
細(xì)胞間粘附分子-1(Intercellular adhesion molecule-1,ICAM-1),也叫做CD54,是一個普遍存在的跨膜糖蛋白,在白細(xì)胞的遷移和激活中起著非常重要的作用??扇艿腎CAM-1被報道出現(xiàn)在血清、腦脊髓液、尿液和支氣管肺泡的洗出液中。在這些液體中 sICAM-1(Soluble ICAM-1)量的提高與心血管疾病、II型糖尿病、器官移植失功、氧化劑應(yīng)激、腹部脂肪聚集、高血壓、肝病和一些惡性腫瘤密切相關(guān)。sICAM-1促進(jìn)血管生成并且可以作為血管內(nèi)皮細(xì)胞激活和受損的一個指標(biāo)。sICAM-1也可以作為跨膜ICAM-1介導(dǎo)活性的抑制劑,比如抑制單核細(xì)胞粘附到激活的內(nèi)皮細(xì)胞,抑制腫瘤細(xì)胞對NK細(xì)胞介導(dǎo)的裂解的敏感性。
產(chǎn)品原理
本實(shí)驗(yàn)采用雙抗體夾心ELISA法??谷薎CAM-1單抗包被于酶標(biāo)板上,同時加入標(biāo)本、標(biāo)準(zhǔn)品和生物素化的抗人ICAM-1抗體,標(biāo)本、標(biāo)準(zhǔn)品中的ICAM-1會與加入于孔中的生物素化的抗人ICAM-1抗體及包被于酶標(biāo)板上的單抗結(jié)合,形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標(biāo)記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有ICAM-1,辣根過氧化物酶會使無色的顯色劑變藍(lán),加終止液變黃。在450nm處測OD值, ICAM-1濃度與OD450值之間呈正相關(guān),可通過繪制標(biāo)準(zhǔn)曲線求出標(biāo)本中ICAM-1濃度。
