產(chǎn)品概述
細(xì)胞白介素2(IL-2)是一種主要由有絲分裂中具有抗原活性的T淋巴細(xì)胞產(chǎn)生的多效應(yīng)細(xì)胞因子。它在促進(jìn)抗原特異性T細(xì)胞克隆中發(fā)揮著重要的作用。另外,已被證明白細(xì)胞介素2(IL-2)可以調(diào)解各種類型細(xì)胞的多種免疫應(yīng)答。白細(xì)胞介素2(IL-2)刺激胸腺細(xì)胞增殖,刺激增殖和分化活化的B細(xì)胞,促進(jìn)生長、分化和滅活單核細(xì)胞,誘導(dǎo)細(xì)胞因子的生產(chǎn)和增長,誘導(dǎo)普通殺傷細(xì)胞增加細(xì)胞溶解活動(dòng),提高淋巴細(xì)胞激活的殺傷(LAK細(xì)胞)細(xì)胞,誘導(dǎo)增殖和分化少突膠質(zhì)細(xì)胞。大鼠白細(xì)胞介素2(IL-2)cDNA為含有155個(gè)氨基酸殘基的前體糖蛋白、包含20個(gè)氨基酸殘基信號(hào)肽,粘合形成成熟蛋白。在氨基酸序列水平,成熟的大鼠白細(xì)胞介素2(IL-2)與小鼠白細(xì)胞介素2(IL-2)的同源性達(dá)80%。
產(chǎn)品原理
本實(shí)驗(yàn)采用雙抗體夾心ELISA法??勾笫驣L-2單抗包被于酶標(biāo)板上,同時(shí)加入標(biāo)本、標(biāo)準(zhǔn)品和生物素化的抗大鼠IL-2抗體,標(biāo)本、標(biāo)準(zhǔn)品中的IL-2會(huì)與加入于孔中的生物素化的抗大鼠IL-2抗體及包被于酶標(biāo)板上的單抗結(jié)合,形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標(biāo)記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有IL-2,辣根過氧化物酶會(huì)使無色的顯色劑變藍(lán),加終止液變黃。在450nm處測(cè)OD值,IL-2濃度與OD450值之間呈正相關(guān),可通過繪制標(biāo)準(zhǔn)曲線求出標(biāo)本中IL-2濃度。
