產(chǎn)品概述
粒細胞巨噬細胞集落刺激因子(GM-CSF)是一個多效性的細胞因子,對造血細胞有多方面的影響。它可以將CD34+前體細胞動員到外圍從而促進它們增殖、存活以及分化成中性粒細胞、單核細胞/巨噬細胞、嗜酸性細胞和骨髓樹突狀細胞。GM-CSF對這些終末分化的骨髓細胞效應(yīng)功能的誘導(dǎo)也是必需的。此外,GM-CSF對于紅細胞和巨核細胞的增殖和分化也起到促進作用。GM-CSF由一系列不同的細胞產(chǎn)生,包括角化細胞、成熟和未成熟的NK細胞、II型肺泡上皮細胞、內(nèi)皮細胞、單核細胞、骨髓間葉干細胞,CD4+和CD8+T細胞、巨核細胞、B細胞、嗜酸性細胞、軟骨細胞和纖維原細胞。
產(chǎn)品原理
本實驗采用雙抗體夾心ELISA法。抗人GM-CSF單抗包被于酶標板上,標本和標準品中的GM-CSF會與單抗結(jié)合形成免疫復(fù)合物,游離的成分被洗去;加入生物素化的抗人GM-CSF抗體,生物素化抗人GM-CSF抗體與酶標板上結(jié)合的標準品或樣本中的GM-CSF結(jié)合而形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有GM-CSF,辣根過氧化物酶會使無色的顯色劑變藍,加終止液變黃。在450nm處測OD值,GM-CSF濃度與OD450值之間呈正相關(guān),可通過繪制標準曲線求出標本中GM-CSF濃度。
