產品概述
白細胞介素17(也稱 CTLA-8)是T細胞表達的一種多效性細胞因子。IL-17和IL-17受體(IL-17R)結合,IL-17R不和任何已知的受體家族的成員具有同源性。IL-17的表達受活化的T細胞的限制, IL-17R 的mRNA廣泛分布在組織中,在已檢測過的所有細胞和組織中均檢測到IL-17R的mRNA。人的IL-17R 的基因位于22號染色體上,它是由866個氨基酸組成的I型膜糖蛋白,包括293個氨基酸組成胞外結構域、21個氨基酸組成的跨膜結構域和一個525個氨基酸組成的胞質尾。人IL-17R的氨基酸序列和小鼠的IL-17R具有69%的同源性。
產品原理
本實驗采用雙抗體夾心ELISA法。抗人IL-17R單抗包被于酶標板上,標本和標準品中的IL-17R會與單抗結合形成免疫復合物,游離的成分被洗去;加入生物素化的抗人IL-17R抗體,生物素化抗人IL-17R抗體與酶標板上結合的標準品或樣本中的IL-17R結合而形成免疫復合物,游離的成分被洗去;加入辣根過氧化物酶標記的親合素,親合素與生物素特異性結合,游離的成分被洗去。加入顯色底物(顯色劑),若反應孔中有IL-17R,辣根過氧化物酶會使無色的顯色劑變藍,加終止液變黃。在450nm處測OD值, IL-17R濃度與OD450值之間呈正相關,可通過繪制標準曲線求出標本中IL-17R濃度。
